How does the AI know when after-hours cover begins?

It answers whatever reaches the Twilio number. The forwarding on your own line decides when that happens: by time of day if your phone system or provider supports it, or switched on by hand when the last person leaves. During the day the line rings at your office as usual.

Can weekend callers hear something different from weeknight callers?

The same AI Instructions apply at every hour, so write them to hold for both. List the full opening times, say that callbacks start at opening time on the next working day, and keep the urgent rule the same. A Saturday caller and a Tuesday caller then each hear an accurate answer.

How do we set up holiday hours?

Add one AI Instruction with the closed days and the reopening time, a few days before the holiday. It takes effect on the next call. Extend your line's forwarding so the daytime hours of the holiday also go to the Twilio number, and delete the instruction when the office opens again.
